RESEARCH EXPERIENCE (Cont'd) Justin
Mahoney /2
Carleton University, Ottawa, ON 2002-2005
Master of Science/Molecular Biology
. Maintained the wild-type model plant, Arabidopsis thaliana by
extracting RNA from the roots, stems, leaves and flowers and performed
Northern blot experiments with the extracts to determine the degree of
expression of the ethylene receptor genes ETR1, ETR2, ERS1, ERS2 and
EIN4
. Extracted DNA and RNA from wild-type, ETR1 and ERS1 mutant plants to
clone various domains of the ERS1 and ETR1 genes
. Transformed wild-type plants with the novel chimeric gene by means of
the "floral dip" method
. Examined, recorded, analyzed and presented phenotypic changes that
resulted from the transformation
. Produced and dissertated thesis, searched bioinformatics, designed
primer, isolated RNA, cDNA synthesis, RT-PCR, cloned genes, gel
electrophoresis, purified DNA, digested DNA, transformed bacterial,
collected and analyzed data, grew and maintained bacterial colony,
transformed "floral dip" plant technique and maintained plant
Accomplishments:
. Created a novel chimeric gene composed of the ethylene-binding domain
of ETR1 and the histidine kinase domain of ERS1; cloned gene used for
further understanding of ethylene signaling pathway
