Guidance

The main objective of the project was to produce single chain antibodies to detect Downy

Mildew through Phage Display Technology, screening of transgenic plants, cloning of TLCV

Replicase gene in to puc-18 vector

Work done,

The polyclonal antibodies against downey mildew of grapes were generated using phage

display library by biopanning procedure.

Fifty monoclonal antibodies specifically binding to pure antigen was screened by ELISA.

Later out of these fifty monoclonal antibodies six antibodies were screened having high

affinity to infected leaf samples.

The monoclonal antibody gene having higher affinity to infected tissue was sub cloned

into alkaline phosphatase construct (pHOA)

The specificity of the construct (pAV Mab-Alp) was confirmed by DAS-ELISA.

The Dip-stick protocol was developed using the above construct as secondary antibody to

detect the infection in specific crop.
