• Performed extensive work on transgenic tobacco plants

• Grew transgenic plants in greenhouse for transgene analysis and protein purification

• Extracted and purified human protein from transgenic plants

• Used different molecular techniques to confirm the presence of transgenes, to detect gene

expression levels and quantify accumulated protein levels

Summary of MS thesis:

Thesis Title: To improve the production of human asialo-erythropoietin (asialo-hEPO) in transgenic

tobacco plants

The aim of my thesis work is to improve the expression level of human asialo-erythropoietin (asialo-

hEPO) in transgenic tobacco plants. To achieve this, a genetic cassette containing human β1, 4-

galactosyltransferase gene (GalT) driven by GapC promoter and erythropoietin gene (EPO) by double

CaMV35S promoter was designed. EPO was engineered to have a Strep II tag and an endoplasmic

reticulum (ER) retention signal KDEL at the C-terminus to aid in the purification (Strep II tag) and

accumulation of higher levels of asialo-EPO in the ER (KDEL). Agrobacterium-mediated transformation

approach was employed to transfer an EPO + GalT genetic cassette into tobacco plants. Transgenic plants

were confirmed for the presence of EPO and GalT genes by PCR. Gene expression levels were measured

by RT-PCR and qRT-PCR. The protein product was detected by ELISA and western blotting. Future work

will purify more protein for its function studies and glycan mapping.
