PhD Dissertation: Investigation of permeability of different chemical form

of an anticancer drug and optimization of transmucosal delivery. The

research involves developing an oral formulation including the following:

. Preformulation- pKa determination, pH Partition studies, pH solubility

studies, kinetics studies at pH 7.4. Equilibrium hydrolysis at

different pH.

. Formulation- In vitro diffusion and transport studies of Drug X through

cellulose, silicone, porcine skin and buccal epithelium. Effect of pH,

concentration and efflux transporters on mode of transport of drug.

Studying effects of paracellular and transcellular enhancers on the

permeability of Drug X through different membranes. Effect of current

on the transport of drug .Films casting using various polymers.

. PK studies- Studying the effect of drug X in rats and rabbits through

IV, IP, sublingual,buccal and oral dosing. Evaluating Pharmacokinetic

profile using WinNonlin, Phoenix. Development of bio analytic method

for plasma quantification LC-MS/MS based bioanalysis, in-vitro ADME

methodologies, metabolite identification and/or PK interpretation.

Comparing Bioavailability through different routes.Extensive hands on

LC-MS method development.

In Vitro Permeation of ON 01210.Na (Ex-RADTM) Through Porcine Skin: This

research was a part of ongoing efforts to develop a readily deployable

formulation for ON01210.Na to be used in a radiation emergency.

Developed /Validated HPLC method for ON 01210.Na.

Conducted solubility and partition coefficient studies for preformulation

characterization .

Investigation of dominant transdermal pathway of drug. Study effect of

various penetration enhancers on permeation of ON01210.Na. Steady state

flux as a function of concentration at physiogical pH.

Correlate the invitro Flux parameters to plasma concentrations.

Master's Dissertation: "Improving drug release of Lidocaine with less

amount of drug in it for lesser toxic effect & economical"

Formulated various ointment bases with or without additives.
