Project 1: Developing a reagent used as reference to calibrate and align the violet laser source, optics and flow rate of a Flow Cytometer.
● Designed Flow Process Map for protocol in order to determine possible control and noise factors in the experimental procedure.
● Identifying prospective dyes for reagent formulation and conducting spectral measurements to observe absorption and emission spectra.
● Conducting Flow cytometry studies using the Attune® in order to gather necessary data.
● Performed Measurement systems analysis(MSA) in order to test user to user, user to instrument and user to sample variability.
● Full Factorial Design of Experiments(DOE) conducted in order to determine ideal concentration of dye and organic solvent in flow reagent.
● Confocal microscopy to determine whether the reagent is violet laser excitable or not.
● Developed a protocol for full scale production of the reagent.
Project 2: Developing small fluorescent amine functionalized hydrogel beads for cell analysis by flow cytometry.
● Attempts to conjugate secondary antibodies(Goat Anti–Mouse IgG) on the surface of fluorescent dye hydrogels using SMCC conjugation method.
● Obtained a Dye vs intensity curve using Attune® Flow cytometer for different hydrogel samples.
● Performed Immunosorbent Assay(ISA) to determine whether successful secondary antibody conjugation took place.
● Imaging of fluorescently stained hydrogel beads using Upright microscope.

PROJECTS:
Synthesis of Bovine serum albumin (BSA) loaded PNIPA PEGDA nanoparticles for wound healing purposes [Spring 2012]
● Three separate batches of PNIPA PEGDA, PEGDA and PNIPA nanoparticles were synthesized.
● Cumulative drug release studies of three batches were done at temperatures above and below lower critical solution temperature (LCST) i.e. at 25
and 37 degrees C.
Synthesis and Mechanical testing studies on PEGMC/methyl diethanolamine (MDEA) hydrogels [Fall 2011]
● Preparation of four different batches of PEGMC hydrogels with varying amounts of methyl diethanolamine (MDEA) crosslinker used.
● Mechanical compression studies of the hydrogels prepared in order to measure modulus of elasticity.
● Nuclear Magnetic Resonance (NMR) and Fourier transform infrared spectroscopy (FTIR) spectra of PEGMC/MDEA.
● Measurement of viscosity of PEGMC/MDEA polymer using both absolute and relative viscometry.
● Measurement of injection pressure and shear thinning studies on the hydrogel prepared.
ACHIEVEMENTS:
● Presented a poster titled ‘Biocomposite Porous Scaffolds for Bone Tissue Engineering Applications’ at the 2012 Materials Research
Society(MRS) Fall meeting held at Boston, Massachusetts, USA
