Management experience:
- Leading a team of 5 to 10 scientists (from technical staff to
experienced PhDs) for delivery of key subcontracting results to
larger pharmas.
- Dealing with customers for project definition and negotiation of
deliverables, timelines and prices.
- Managing reporting and communication to customers and management.
- Safety issues (incl. radiotracers).
- Lab budget supervision and cost cutting.
- EU funding applications and French tax credit.
- Setting up and running collaborations with academic teams.
Field of Expertise: Design, production and assay of proteins in the drug
development process.
. Design of protein constructs for structure determination or HTP assays
:
Transforming the complex drug targets identified by cellular biology
to into constructs suitable for production, drug development and
screening:
. Development of banks of antibody like binder-proteins (Scfv and
original scaffolds).
- Validation of the scaffolds.
- Display and screening methods.
- Binder validation.
. Production :
Design and implementation of dependable protein production processes:
- Expression (E.Coli to mammalian cells).
- Purification : analytical scale to upstream production process
from recombinant or natural proteins.
- Protein refolding.
. Structure determination :
Whole process from construct to 3d structure (x-ray or NMR) applied to
drug discovery with a focus on the identification of the drug binding
sites.
. Protein QC:
Ensuring the suitability of the constructs for DD or structural use:
- Biochemical assays and validation.
- Biophysical methods : Mass spectrometry, Electrophoresis (incl 2d),
Thermofluorimetry, Light scattering DLS MALS, angle x-ray (or
neutron) scattering, NMR.
. Design and validation of activity assays in-vitro :
- Setup of assays using radiotracers or absorbance or fluorescence;
(ex Gef proteins, Kinases). Ubiquitination assays. Fluorescence
polarization.
- Setup and transfer to customer of a luminescence assay to replace
radioactive assays; (e.g. t-RNA synthetase assay for the screening
of natural antibiotic compounds).
