Complete a project on clinical utility of TaqMan probe in quantification of hepatitis B virus by Real Time PCR.
DESCRIPTION
Hepatitis B is an infectious disease caused by the hepatitis B virus (HBV) which affects the liver. It can cause both acute and chronic infections. Many people have no symptoms during the initial infection. Some develop a rapid onset of sickness with vomiting, yellowish skin, feeling tired, dark urine and abdominal pain. HBV has eight genotypes (A-H) due to variation in nucleotide sequences greater than 8%. Various publications are found that the clinical course and outcome of antiviral therapy dependent on the genotype of the infecting HBV strains. After that real time come in field of molecular diagnostic of epidemic diseases. The Roche Real Time PCR accurately identifies the genotype in one step reaction by means of primer specificity. It provides a useful tool for rapid detection and would make large scale longitudinal HBV related studies simple & feasible. TaqMan hepatitis B virus (HBV) analyse specific reagent is designed for the quantification of HBV DNA in serum or plasma. Analytical sensitivity and precision were assessed with commercially available HBV standards, while clinical serum specimens from HBsAg seropositive patient and healthy blood donor were used to determine clinical sensitivity, specificity and correlation with other commercially available assays

Characterization of Gut Microflora of Humans (Homo sapiens) Living in the Tribal Areas of Himachal Pradesh.
DISCRIPTION
The relevance of the gut microbiota to human health is increasingly appreciated. The objective of this study was characterization of the gut microbiota of a group of teenage and children living in tribal areas of Kinnaur district of Himachal pradesh, India. Faeces were collected from 10 healthy tribal teenagers and child of 1- 25 years from the Karchham village of Kinnaur district, Himachal pradesh. DNA was extracted, and amplified by using 16s rRNA universal bacterial primers and they showed approx. 1500bp segments comprising region of 16S rRNA gene were amplified, barcoded and sequenced. Totally 600 and 750 base pairs good-quality reads were analyzed. Tribal peoples consumed a millets-based and rice-based diet, ate cereals and traditional Himachali fermented food every day, and consume milk or milk products. Meat once a week. In both groups, Escherichia was the most abundant phylum, followed by various uncultured bacterial clones. Phylogenetic analysis of major microbiota indicated clustering of microbial groups at different major branch points for tribes. Genus Escherichia and various uncultured bacterium clones constituted the bulk of the faecal microbiota, while significant differences in composition between the groups were probably due to differences in diet and lifestyle and age factor. 1) IS6110 Gene in Mycobacterium Tuberculosis Complex in Tuberculosis Meningitis- Clinical Application for Disease Monitoring (Scholars Academic Journal of Biosciences) Tuberculosis meningitis (TBM) is the most common form of chronic infection of central nervous system. Regardless of the extent of the problem, the general diagnostic outlook is discouraging specifically; there is no generally accepted early confirmative diagnostic protocol available for TBM. In the current work, 16 Cerebrospinal Fluid (CSF) cases were considered for the molecular characterization.02 CSF came positive for the Nested PCR targeting IS6110 gene, considered to be positive for the presence of mycobacterium tuberculosis DNA. It was also seen that the positive cases were with increased ADA titer. The clinical manifestations of the positive cases were matching with those for the tuberculosis meningitis. 2) High-Risk Human Papilloma Virus (HR-HPV) Genotyping in Female with Abnormal Cervix

Human Papilloma Virus (HPV) infection is transmitted primarily via sexual and affects mostly basal epithelial cells of the skin or inner lining of tissues and are categorized as cutaneous types or mucosal types. It is known that HPV infection is strongly related to the development of cervical cancer, as viral DNA is detected in 99% of cervical cancer cases worldwide. Proposed study includes 14 cervical brushing, collected from females with cytologically abnormal cervix. The target genes E6 and E7 were amplified by Conventional PCR. Three cases with High grade squamous intraepithelial lesion (HSIL) came positive. Further the positive cases were digested by restriction endonucleases enzymes for RFLP and the amplicon size for high risk HPV type 16 and type 18 were obtained at 452bp and 455bp respectively. Three cases came positive, out of which HPV type 16 came in two cases with one HPV type 18. High risk HPV characterization at an early stage will prevent further progression of the disease.
