SPECIALISATION: Microbial Biochemistry.

THESIS TITLE: “Biofuel Production from Marine Phototrophic bacteria” Supervisor: Dr.M.P.Pratap Rudra, Department of Biochemistry, Osmania University. Brief Outlines of Research Work:

Collection of different marine water samples from Kanyakumari (Indian Ocean), Vizag

(Bay of Bengal), and Goa (Arabian Sea).

Isolation of organism (Purple non-sulphur bacteria or an oxygenic Phototrophic bacterium) from different marine samples.

Characterization of organism by 16s rRNA sequencing for identification of strain.

Deposition of identified organism to NCBI for specific identified strain for my isolated cultures.

Optimization studies of Different carbon, nitrogen, growth factors and PH variation were checked by identified an oxygenic phototrophic bacteria and their growth variation in different parameters.

Biochemical Characterizations like Carbohydrate, Protein, Cellular carbon & nitrogen ratio, bacterial pigments, total amino acids and also indole acetic acid estimations were done for identified isolated organisms from different marine water samples.

For all the above parameters which were checked for Hydrogen gas production by water displacement method and also by Gas Chromatography.

The isolated strains were expressed for cloning, gene expression and characterization of aminolevulinic acid synthase and also the shown the variation of the hydrogen gas production after the expression studies.

Anticancer studies by MTT assay and flow cytometric apoptosis analysis for the isolated bacteria against the breast cancer cell lines SKBR3 and MDA-MB231. Laboratory Skills developed during my research project:

Microbial techniques like isolation of bacteria by culturing in the enriched media and by spreading, pouring by Para film overlay method, gram staining.

Biochemical techniques for the carbohydrates, protein and indole acetic acid and also the total amino acid content in the isolated bacteria by using HPLC.

Molecular biology and rDNA technology- DNA isolation, SDS-PAGE, Restriction digestion, PCR, Sequencing, cloning, transformation, agarose gel electrophoresis, and MTT assay and cell culture studies, etc.

Bioinformatics- BLAST,FASTA and Soft wares like MEGA 7,Origin for statistical analysis and graphical representation and NCBI deposition of the identified strains for the obtaining the accession numbers.

Special Skills: Isolation and synthesis of nanoparticles from the various plant sources and published the publications on the synthesized nanoparticles. WORK EXPERIENCE:

Worked as a lab technician in Sree Tirumala Medicare and DRC PVT.LTD, Kalyani Hospital, Hanamkonda from 2-8-2007 to 16-7-2009.
