Summary of Ph.D. thesis work
Plectranthus bourneae is an endemic plant, belongs to the family Lamiaceae. It is distributed at Pambar Shola, Palni hills, Kodaikanal, Tamil Nadu, India. Matthew (1999) reported the presence of this endemic species in Pambar Shola of the Palni Hills, Tamil Nadu, India, since then it has been a rare plant to collect due to loss of habitat by anthropogenic activity. The present study deals with phenology, seed germination and standardization of in vitro protocols for germplasm conservation. The phenology of this plant provides monthly events of vegetative, flowering and seed set of P. bourneae at Palni hills. Micropropagation was achieved through solid and liquid medium from axillary bud and shoot tip explants. Of the explants, axillary bud explants in liquid medium produced the highest average of 21.42 shoots/explant on BA 0.7 mg/l + TDZ 1.0 mg/l + AdS 50 mg/l. Organogenesis was achieved using leaf, internode and petiole explants through direct and indirect phase. Among the two-different phases, indirect organogenesis developed highest number of 29.71 shoots from leaf explants. Rooting was best achieved on IBA 1.0 mg/l with an average number of 19.14roots/shoot. Somatic embryogenesis was achieved through direct and indirect phases. Embryogenic callus followed by successive stages of (globular, heart-shaped and torpedo-like) somatic embryos formed from leaf and internode explants. The genetic fidelity of plantlets obtained from micropropagation, organogenesis and somatic embryogenesis were cross checked against their respective mother plants using ISSR markers. Antibacterial activity of various solvent extracts (leaf and stem of P. bourneae) was studied for various Gram positive and negative bacteria. Of this ethanolic extract was found to be most potent, therefore the leaf and stem ethanolic extracts were subjected to GC-MS and several potential antibacterial compounds such as alpha.-Amyrin, n-Hexadecanoic acid, Palmitic Acid, and Stigmasterol were identified.
