Anti-inflammatory and anti-tumor function of spasmolytic polypeptide/trefoil factor 2 (SP/Tff2) in colon and gastric cancer.
Responsibilities:
1.Design, plan and execute experiments to examine the role of TFF2 in gastric and colon cancer by using following techniques:
Mouse models: DSS-induced colitis, gastric, colon and liver cancer, maintaining mice colonies, genotyping
FACS analysis of immune cells populations from mouse immune, gastric and colon tumor tissues
phenotypic and functional characterization of myeloid-derived suppressive cells (arginase-, ROS- activity, immunosuppressive activity)
isolation myeloid and T-cells from spleen and bone marrow for in vitro culture and adoptive cells transfer via tail vein injection
Purification and functional characterization of recombinant mouse TFF2 and its derivates in tests in vitro and in vivo in mouse cancer models
Adenovirus-mediated gene therapy: adenovirus production, purification and tail vein injection
Western blotting, PCR, RT-PCR
Immunochistochemical staining of frozen tissue sections
Mouse bone marrow transplantation and analysis of mouse chimeras
Computer skills: Microsoft Word, Adobe Photoshop, Power Point, Excel, GraphPadPrism
