Research Experience: 4 years

1) Worked as research student under the guidance of Prof. Bengt Hallberg,

Department of Molecular biology, Umea University, Sweden.

Worked on Investigation of function of anaplastic lymphoma kinase (ALK) and its

downstream signaling, mutants of ALK gene have been identified in neuroblastoma

patients, the occurrence of these mutations are noted and transiently transfected mutants

are constructed in different cell lines. These cell line models have been studied for

checking the characteristic futures of neurite formation; effect of mutations on

proliferation, cell signaling and downstream events of ALK activation using some

inhibitors is also studied. ALK expression patterns and its signaling analyzed using

western blot, ECL development and chemiluminescence.

2) Teaching experience:

3) Work experience:

Worked as a Junior Research Associate for Molecular Connections,

Basavanagudi, Bangalore, India.

Mining of literature to construct signaling cascades with genes, proteins and its

interacting small molecules, this data was compiled to build a scientific database.

Expertise in Bio-curation, Protein expression profiling and epigenetic aspects of

genes.Worked on Literature informatics aspects of proteases and protein substrates.

Excellent in understanding biological research papers, reviews and journals. Experience

in 3 different projects related with Diabetes and Cancer and have had many opportunities

4) Project title: “Oxidative modification of high density lipoprotein associated

Paraxonase: Role of water and fat soluble antioxidants” under the guidance of

Prof.Cletus.J.M.D.Souza, Department of studies in Biochemistry, Manasagangotri,

University of Mysore, Mysore

Paraoxanase enzyme was obtained from human serum and purified using cibacron gel, its

activity was tested by PON assays spectrophotometrically by time kinetics. As this

enzyme is associated with HDL we tested its oxidation using H2O2, lipid peroxides and

copper catalyst. Further the Oxidation of PON by H 2O2 and copper was inhibited by

water soluble antioxidants like ascorbic acid and analog of curcumin, and lipid peroxide

caused damage of PON was recovered by fat soluble antioxidant curcumin.

We derviatized Curcumin by attaching glycine this became water soluble and was an

antioxidant to water soluble oxidants like Cu++ and H2O2.

5) I worked as a Research student on “Action of garlic on lipid peroxidation and

antioxidation enzymes seen in the animal models having skin carcinoma induced by

7,12-dimethylbenz[a]anthracene”. In Department of studies in Biochemistry,

University of Mysore, Mysore, Karnataka, India.

I worked on the action of dietary phytochemicals such as garlic having cancer

protective effects, we introduced skin carcinoma by 7,12-dimethylbenz[a]anthracene in

swiss albino mice model, pre and post garlic treatments where given to different groups

of mice and now we are investigating the histology of the affected skin. Along with that

we performed biochemical assays using the liver tissues of mice, for lipid peroxide by

TBA assay, catalase, superoxide dismutase by pyrogallol auto-oxidation and glutathione

peroxidase spectrophotometrically which demonstrate the effect of garlic in mice. Further

results obtained, proves that the Garlic induces elevation in levels of antioxidants, which

was accompanied by decreased lipid peroxidation and incidences of papillomas, thus

protecting cells from injury by the oxidized molecules.

6) Project title: “Over expression and purification of GST-RdRp” under the guidance

of Prof. H.S. Savithri, Dept of Biochemistry, Indian Institute of Science (IISc),

Bangalore.

The RdRp clones 52GST RdRp,62GST RdRp,52CΔ43GST RdRp and 62CΔ43GST

RdRp were transformed into BL21 Rill, BL21 Rosetta (DE3),BL21 pLys S cells and BL21

C43(DE3) competent cells incubated and heat shocked for 5 minutes and later plated on

LB-amp plate, positive cultures were obtained in large quantities using LB amp broth

(Lauryl tryptose broth). Culture was induced with IPTG and incubated, sonicated,

centrifuged, supernatant put for GST binding and protein eluted using GST elution buffer.

The eluted protein samples were loaded on SDS-PAGE and western blotting was done.

Protein was further dialyzed and used for characterization purpose.

Duration: 3 months (Summer project 2006)
