> Worked in Defence Research and Development Establishment (DRDE) as a
Trainee for the accomplishment of M.Pharm. dessertation on
"immunological and toxicological studies of abrin isolated from abrus
precatorius"
o The work requires me to use basic techniques like Delivery of dosage
form to animal, dissection of animal, Calculation of dosage to be
delivered, centrifugation, Determination of LD50 of toxin, & advanced
techniques like DNA isolation, development of polyclonal antibody,
purification of antibody, DOT ELISA, 2D Electrophoresis,
Haemagglutination test, Agarose gel electrophoresis, SDS-PAGE,
Oxidative stress test like Glutathione assay by Spectrophotometer,
Lipid peroxidation test by Fluorimeter and Bioplex cytokine assay by
Bio-PlexTM200; and biochemical parameter detection by Erba-chem
analyser.
o I learnt how to execute and how to plan an experiment, and by working
on these techniques and applying my knowledge practically gave me the
confidence to apply my skills to other projects as well.
> In B.Pharm worked on " Standardization of marketed ayurvedic products-
Abhyarishta" and " Documentation in Pharmaceutical Industry as per
GMP"
o Through the work I learnt methods which are used for the
standardisation of ayurvedic preparation and these methods evidently
showed the difference in different marketed products which showed
their authenticity.
o I also learnt what the prescribed documentation requirement is in the
pharmaceutical industry as per GMP.
Technical Expertise:
I have technical knowledge in:
> Genomic DNA isolation from bacteria & plant material.
> Agarose gel electrophoresis & SDS-PAGE with the help of BioRad
instrument.
> Isolation of protein from liver tissue, serum, plant seeds and its
quantification by Bradford Method.
> Operating of UV-Spectrophotometer (Perkin, CS-55, UK)
> HPLC equipments in analyzing the biological samples. (Shimadzu SPD-28)
> Culture Techniques- Spreading, Streaking, Pour plate technique.
> GC equipments in analyzing sample ( chemito,perkin Elmer)
> Karl Fischer apparatus for water content in sample.
> Aseptic Techniques
> Plant Tissue Culture - Samples used were a apical stem, leaf and a
seed which were grown on solidified agar medium under aseptic
conditions.
> DOT ELISA - Samples were different dilutions of purified antibody
developed in rabbit against toxin to check the titre.
> Western Blotting - Samples were purified abrin and polyclonal antibody
raised in rabbit.
> 2-D Electrophoresis - IPG strip (7 cm, pH 3-10), IEF unit (Bio-Rad,
USA).
> Histopathology - Samples were abrin treated swiss mice liver, spleen,
heart, kidney, brain; Equipment used were Microtome(Shandon Finesse),
Tissue Embeding Machine (Yarco Scientific), Tissue Processor (Yarco
Scientific).
> Haemagglutination assay- Lin and Li, 1980
> Cytokine assay- Bio-PlexTM200 (Luminex xMAPTM Technology)
> Chromatographic purification using protein purification system, Bio-
Rad, USA (Biologic-HR model)

. I have been a part of guest seminars by great personalities like Alec
Griffey's, Venkatraman, and Werner Arbor in M.Pharm. where they
delivered a very informative/exciting/conceptual lecture/talk on DNA
Fingerprinting, Structure and function of Ribosomes and importance of
RNA, role of restriction enzymes respectively.
. I have attended inspirational guest seminars in Annual National
Biotechnology Festival i.e., INCYTE-2011(18-20th January). In this I
learnt about-

- Protein Folding Paradox and what is India's stand on Bioinformatics

. I have attended seminar on "Self medication by Pharmacist Safe or
unsafe" in B.Pharm.(2006) which was the first lecture I attended and
that made me feel the need of pharmacists and his/her role in society
and how at large it is being neglected.
