Identification of host cell factors that are involved in the replication of Adeno-

associated virus (AAV). This has involved extensive cell fractionation and protein

purification as well as recombinant protein work to identify the essential proteins

required for virus replication.

Design of AAV gene therapy vectors for the study and treatment of Alzheimer s

disease. To this end we have produced recombinant AAV, that in preliminary

studies, reduces the -amyloid load on Alzheimer model mice. We are also

examining the use of AAV with shRNA to reduce the levels of BACE protein in the

same animal models.
