Method Development for Analysis of Testosterone / Epitestosterone in
Human Samples Using LC/MS/MS
Worked on a separation method-development of
testosterone/epitestosterone using LC/MS/MS (Thermo TSQ Quantum with
triple quadrupoles, mostly on ESI, some APCI mode), software:
Xcalibur 2.0.6
10 pg/mL LOD and 50 pg/mL LOQ of standard solutions and reconstituted
urine samples were targeted for a faster and more sensitive method
than existing GC/MS/MS methods utilizing derivatization of
sugar-conjugate steroids
Urine sample preparation: enzymatic hydrolysis of steroid conjugates
followed by solid supported liquid-liquid extraction (Hydromatrix)
/SPE (Isolute C8)
Trouble shooting on HPLC and ESI sprayhead, cleaning the capillary
inlet, replacing tubing, fixing clogging, assisting the supervisor
cleaning quadrupoles
Data collection: exporting and organizing data onto excel sheets
Good analytical laboratory practice: accurate and precise
measurements, paying attention on details, good notebook keeping
