A single quantum dot-smFRET based bionano-sensor to monitor the
proteolytic activity of matrix metalloproteinase (MMP 14) on QD-peptide
bioconjugates. For this we used the principles of single molecule
Fluorescence Resonance Energy Transfer (smFRET) and Total Internal
reflection fluorescence microscopy (TIRFM) along with biocompatible, water
soluble quantum dots (CdSe-ZnS) as fluorescent probes for the single
molecule detection and imaging.
Polymeric colloidal handles incorporating biocompatible, water soluble
QDs by layer-by layer self assembly and ligand exchange processes. These
micron sized particles have applications in cellular targeting and imaging,
colloidal handles to carryout force measurements by optical trapping and as
fluorescent probes for experimental fluid mechanics.

Development of a rapid and early analyzer using the magnetic tweezer
setup for the detection of heart attacks in conjunction with Sensera Inc.
Boston. This project involved fabrication, physical characterization and
surface functionalization of porous alumina membranes for picogram level
detection of cardiac troponin in blood serum. Also surface functionalized
superparamagnetic iron oxide microparticles were used for detection of
cardiac troponin in blood.

MS Thesis: DNA containing Layer-by-Layer thin films on colloidal
microparticles for gene delivery
Development of a microcapsule based DNA immunization system using the
layer-by-layer self assembly process. System consisted of biodegradable
polymeric microparticles with layers of poly (ethylene mine) and plasmid
DNA. Physical Characterization using zeta Dynamic Light Scattering and
scanning probe microscopy was achieved. Studies involving transfection and
cellular uptake (using Flow cytometry) of the microcapsules by macrophage
and dendritic cell lines were carried out. HPLC was used to study the
release profile of proteins and drugs from these microcapsules.
Development of an ultrasound assisted perfluorocarbon emulsion droplet
based system, containing polyelectrolyte shells for cancer immunogene
therapy.
