. Performed Molecular techniques such as Isolation of genomic
DNA, Plasmids, and RNA, PCR, Real-time PCR, Cycle Sequencing,
TOPO cloning, Gateway recombination and transformations,
quantified nucleic acids using nanodrop using pico green and

Thesis: Characterization of the promoter of a Nutrient-stress
Responsive Gene in white lupin
. Performed primer-walk sequencing to obtain the putative
promoter of L#315 a nutrient stress responsive gene.
. Performed TOPO cloning and Gateway recombination techniques
to do transcriptional fusion of L#315 promoter with GUS and
GFP reporter genes independently and transformed the
constructs into white lupin plants.
. Performed Real-time PCR to qualitatively analyze the
presence of GUS and GFP reporter gene in the proteoid roots
of the transformed plants. Also compared the expression
level of the gene of interest in nutrient stress and
nutrient sufficient condition.
. Used Agarose gel electrophoresis to confirm the PCR
products.
. Used histochemical assay to visualize GUS expression &
Fluorescence Microscope to observe GFP expression in the
proteoid roots of the transformed plants.
