Prepared Hepatopac plates using different donors of hepatocyte
Media preparation, freeze thawing, shake flask, and maintenance of primary hepatocyte culture.
Cultured the 96 well micropatterned plates with primary hepatocyte cells and fibroblasts, and
maintained the cells.
Dosed the cells with different inducers and repressors of cytochrome-p450 (CYP3A4) and
performed functional characterization like CYP3A4 promega glo P450 assay, albumin secretion
assay, and urea synthesis assay.
Transporter Characterization in HepatopacTM platform.
Uptake and efflux study of 4 different drug compounds on HepatopacTM cell culture platform.
Perform RT-PCR based gene expression of 6 different genes (NTCP, OCT1, MRP5, MDR1, BSEP, Actin)
coding for a particular transporter molecule. Performed functional characterization of HepatopacTM.
Compared the gene expression of HepatopacTM culture platform with the suspension culture.
Study the effect of kupffer cell mediated cell toxicity on HepatopacTM
Co-cultured kupffer cells on HepatopacTM, and studied the effects of two different compounds
diclofenac and troglitazone on HepatopacTM and kupffer cell cultured HepatopacTM.
Performed functional characterization on both the cell types and also performed GSH and ATP
