Research Experience

Post Doctoral

Working on “Discovery of extremozymes from extremophiles isolated from natural resources for

industrial application”. (Jul 2009 to October 2011).

Extremophilic microbes produced extremozymes which have potential use for several industrial applications. Various

extremophiles were isolated from lake, yard waste compost, hot springs, Yellowstone National park, DUSEL.

Extremophiles have potential in cellulase and xylanases production using lignocellulosic substrates for fermentable sugar

production followed by ethanol fermentation. Simultaneous saccharification & fermentation (SSF) and separate hydrolysis

& fermentation (SHF) for biofuel production were carried out with experimental and commercial enzymes. I worked on

biofuel and bioenergy production including- bioethanol, biohydrogen, biodiesel and bioelectricity through microbial fuel

cell. Initiated research work on lichen as a source of microbes for bioactive component and biodiesel production .

Worked on “Strain improvement for extracellular enzyme of various bacteria and fungi using

classical mutation” (Nov. 2007-Jul 2009).

Several extracellular enzymes including amylase, amyloglucosidase, protease, serratiopeptidase, cellulase, xylanases,

lactase and phytase were produced from various bacteria and fungi. Classical mutation by physical and chemical agent

employed for strain improvement of all enzymes from various microbes. Strains were isolated and were studied for several

generations with various cultural and nutritional parameters. Potential enzymes were characterized by HPLC, 2D gel

electrophoresis.

Soil-1 and BSA 26, dehairing protease-producing microorganisms were isolated from vermicompost pit soil and buffalo

hide respectively. Both the cultures were identified according to the Bergey’s Manual of Systematic Bac teriology and by

molecular technique of 16 S rDNA. Optimization of protease production was carried out with different cultural conditions

(aerobic- anaerobic, shake- steady, shaking speed, cultivation volume); environmental parameters (pH, temperature and %

inoculum size); nutritional parameters (carbon, nitrogen sources, inorganic salts, agro products/ byproducts, non ionic

surfactants- detergents, antifoaming agents). A statistical tool of response surface methodology (RSM) was also studied for

optimization of final ingredient concentrations. Production optimization of protease was carried out at 10L fermentor with

respect to agitation and aeration. Scale up of protease production was carried out at 100L fermentor. The protease was

purified by ammonium sulphate precipitation, gel filtration and preparative gel electrophoresis and characterized. The

crude enzyme has potential application in soaking, dehairing and bating operations of leather processing. In addition it has

application in detergent industry. It has extensive application in recovery of silver from waste X- ray and photographic

films.

Lonar lake isolate, Bacillus sphaericus, was tested for protease production followed by media optimization with respect to

various cultural, environmental and nutritional parameters. The protease was purified by ammonium sulphate precipitation,

gel filtration and DEAE sepharose and characterized. The gene responsible for protease production was cloned in E. coli

strain and studied for its expression. The crude enzyme has potential application in detergent industry.

Worked on “Adjuvant therapy of Plumbago zeylanica against UTI microorganisms” (2000- 2001)

M.Sc. part II Dissertation.

Antibiotic resistant pattern is developed in many microbes, especially in enterobacters. Antibiotic resistance pattern was

done. To solve the problems use of plant based natural products will be the best so lution. Plumbagin, a napthquinonic

compound extracted and characterized from plant Plumbago zeylanica. Various extracts were evaluated for antimicrobial

activity against pathogenic bacteria like E. coli, Salmonella sp., Shigella sp., Klaibsiella pneumonae, S. aureus and P.

aeruginosa. Plant extract was used as a curing agent in treatment of infectious diseases at plasmid DNA level.

Industrial

7. Worked on bacterial/fungal fermentation (solid state and submerged state), media designing, process optimization, and

strain improvement using rational, statistical and classical mutational approaches for microbial enzyme production,

fermentation scaled up from flask level to 1000L fermentor . Also as a advisor with one of Indian Agricultural Company,

we developed total 6 products (1-fungicide from Trichoderma viridae, 4-bioinsecticide from Beauveria bassiana,

Verticillium becanii, Metarhizium anisopliae, Pseudomonas fluorescence and 1- nematocide from Paecilomyces

lilancinus).
