The catabolism of branched-chain amino acids (BCAAs) is complex and involves a large number of enzymes. The initial enzymes of the pathway are being studied by our group. To proceed with the investigation, it is highly relevant to study the steps catalyzed by the enzymes Methylglutaconyl-CoA Hydratase and 3-Hydroxyisobutyryl-CoA Hydrolase (specific to the pathway converting Leu to Acetyl-CoA and Acetoacetate). As the final products of this pathway are oxidizable and can function as energy sources or potential substrates for the biosynthesis of fatty acids, we have proposed the hypothesis that these enzymes are relevant to the bioenergetic functions of the parasite. In summary, this project aims to unravel in detail the characterization of enzymes involved in the relationship between BCAAs and their role in the bioenergetics of the parasite. For this, different and multidisciplinary approaches will be used, such as cloning and expression of proteins, classical enzymology, measurements of mitochondrial parameters such as membrane potential and oxygen consumption, and NMR metabolomics, among others. The ideal candidate, should have at least a strong expertise in parasitology (experience in culturing of all life stages of Trypanosoma cruzi is fundamental) and fundamental biochemistry and molecular biology.

Who we are?

Our research group is based at the Department of Parasitology, Institute of Biomedical Sciences at the University of São paulo, in São Paulo – Brazil. Our groups has been working on the discovery of fundamental aspects of trypanosomatids metabolism and bioenergetics, as well as the possible application of the knowledge acquired through our research for the development of new drugs with therapeutic potential for the diseases caused by these pathogens.
