Research Experience

•Structural characterization of Amyloid β peptides Aβ(1-40) and Aβ(1-42) primarily by Solid-State NMR spectroscopy (SSNMR) using magic angle spinning (MAS) and cross-polarization (CP) 1D, 2D homonuclear dipolar recoupling experiments such as RFDR, DARR, heteronuclear double CP experiments and internuclear distance measurements from REDOR, CTfpRFDR experiments

•Molecular level metal (Cu2+/Zn2+) binding structures characterized by CP-MAS 1D, 2D and 13C, 15N and 1H paramagnetic relaxation (T1) experiments. Also used other methods like UV-VIS, fluorescence, circular dichroism and transmission electron microscopy for the characterization purposes

•Sensitivity enhancement in SSNMR by paramagnetic assisted condensed data collection method for nanomole scale protein micro crystals, amyloid fibrils and metallo proteins in hydrated and lyophilized conditions

•Have experience in handing first of its kind 1mm JEOL ultra high speed MAS probes (>70kHz) and 1.8mm high speed MAS probe (40kHz) . Handled 2.5mm and 3.2mm MAS probes frequently for routine analysis. Have knowledge in fixing solids probes.

•Have experience in working on 800MHz, 900MHz Bruker NMR instruments (Solids and Solutions equipped with cryo probe). Gained knowledge in setting up 1H detection experiments at ultra-high speed MAS (>70kHz). Implemented 1H detection pulse sequences with low power heteronuclear decoupling schemes

•Expertized in synthesizing, purifying and analyzing Aβ(1-40) and Aβ(1-42) peptides by solid phase peptide synthesis (SPPS) and in crystallizing proteins for SSNMR analysis using polyethyleneglycol (PEG) and methylpentanediol (MPD) precipitation. Have experience in hanging and sitting drop crystallisation methods

•Handled PC-12 Rat pheochromocytoma cells and performed toxicity assay experiments on these cells

Jan 2003- July 2006

•Structure elucidation of small organic molecules and their impurities in pharmaceutical products formed during drug discovery/process development/formulations/production using solution NMR experiments like DQCOSY, HSQC, HMBC, TOCSY, NOESY, HSQCTOCSY. Have experience in setting up LC-NMR experiments for impurity structural characterization

•Absolute configuration determination of chiral molecules using 1H and 13C solution NMR experiments

•Have good experience in interpretation of IR, UV-VIS, MALDI, electrospray, FT-MS, EI, CI mass spectral data

•Have sound knowledge in the solid-state characterization of drugs using Powder X-Ray diffraction, Thermal analysis Differential Scanning Calorimetry (DSC) and Thermo gravimetry (TG)

•Have grown single crystal x-ray diffraction quality small molecule organic crystals and co-crystals and also have experience in collecting x-ray diffraction data and solving the crystal structures.
