I AM WORKING IN RECOMBINANT PROTEIN PURIFICATION PROCESS LIKE HUMAN INSULIN, GROWTH HORMONE, TRYPSIN AND STREPTOKINASE.

I HAVE EXPERIENCE OF HANDLING THE FOLLOWING UNIT OPERATIONS, WHICH INVOLVES THE PREVENTIVE AND PREDICTIVE MAINTENANCE OF OPERATIONS

• OPERATION OF PILOT SCALE BIO CHROMATOGRAPHY SYSTEM : Akta Pilot-GE Health Care and K-prime 40II,21CFR part II (Millipore)

• COLUMN MAINTENANCE: Packing, Unpacking, Cleaning, Storage and maintenance of Column (HIC, IEX and AP).

• COLUMN WASHING: By NaoH and WFI (Water for Injection).

• COLUMN EFFICIENCY: HETP and Asymmetry

• COLUMN EQUILIBIRATION: By Using Tris and urea Buffer.

• STERILIZATION OF MATERIALS: Hot Air Oven and Horizontal Autoclave.

• SOLUBILIZATION OF PROTEIN: Inclusion bodies (after upstream process) were solubilize by using 8M urea buffer

• CENTRIFUGATION: Solubilized protein were separated by continuous centrifuge (WESTFALIA & ALPHA LAVA) and collection of supernatant for affinity purification

• AFFINITY PURIFICATION: Fraction elution in gradient mode using tris and urea buffers

• REDUCTION AND EXCHANGE: Collection of Affinity purified fraction is subjected to reduction and exchange process with buffers

• REFOLDING AND EXCHANGE: Reduced sample are refolded at 4 C in the presence of refolding buffer

• ENZYME DIGESTION: Refolded sample is subjected to enzyme digestion in the presence of tris buffer and enzyme.

• ION EXCHANGE CHROMATOGRAPHY : Enzyme digested sample loaded to ion exchange column and elute the Fraction in gradient mode using elution buffers

• PILOT SCALE HPLC SYSTEM: Collected ion exchange fraction to be loaded in HPLC and elute the fraction with elution buffers

• EXCHANGE PROCESS: all exchange process is carried out in TFF System (Grandstand Dual Position Membrane System- GE Healthcare).

• CRYSTALLIZATION: HPLC fractions are crystalized zinc crystallization method.

• LYOPHILLIZATION: Zinc crystalized sample are finally lyophilized.

JOB RESPONSIBILITIES

Handling of instruments like HPLC, UV visible spectrophotometer, k-prime, Akta pilot, Akta prime, columns, continuous centrifuge (West Falia and Alpha), etc.

Calibration / maintaining instrument of the production.

Follow the SOPs and cGMP while working on the shop floor. Product batch size selection, Maintains, monitor & control process parameter, filling of the batch tickets (BPR), startup & shutdown of all operations.

Maintain production quality & product yields.

Maintain safety and good housekeeping of work area.

Looking after general administration of laboratory, identifying various chemical reagents, equipment spares and other lab consumables.
