Internship at Department of Biophysics, Panjab University, Chandigarh
Supervisor: Dr. Sarvnarinder kaur
Duration: January to June, 2020 6 Months
Title of internship: To determine the chemopreventive potential of hydro-ethanolic Murayya koenigii leaves extract (HEMKLE) against DMBA/TPA induced skin carcinogenesis in mice. Description: Skin cancer was induced by applying DMBA/TPA on the skin of mice, resulted in DNA mutations occurred in oncogenes and tumor suppressor genes. The observations of the present study clearly indicated that HEMKLE exerted remarkable chemopreventive effects against DMBA/TPA induced skin carcinogenesis. The retardation in the development of skin carcinogenesis upon HEMKLE supplementation was revealed through reduced histo-architectural alterations, reduced incidence as well as total number of tumors. It has also shown to modulate key processes like ROS generation, LPO levels and activity of antioxidant enzymes which are crucial in the initial stages of chemical carcinogenesis.

Internship at Institute of Nano Science and Technology, Mohali
Supervisor: Dr. Manish Singh
Duration: May to July, 2018 2 months
Title of internship: Quantitative analysis of azo dye Eriochrome Black T (EBT) in industrial effluent and its in vitro toxicological studies.
Description: Determination of the quality of water samples
(from industrial effluent sites) by measuring Total dissolved solute (TDS) concentration, COD, and BOD and concentration of EBT in water sample was measured by UV-VIS
Spectrophotometry. MTT assay was performed to measure the cytotoxicity of EBT on NIH-3T3 cell lines, revealing that increase in dose of EBT resulted in increased cell toxicity as well as cell mortality.
Bacterial culturing techniques- Media preparation, stearilization, filtration, spreading, streaking, subculturing, bacterial isolation, preservation, physical and biochemical testing.
Downstream processing- DNA extraction, RNA extraction, centrifugation, protein extraction and purification (filtration, concentration, electrophoresis, chromatography, and dialysis)
Immunology- ELISA, immune-precipitation, immune- agglutination, and Dot Blot
Biochemistry- Qualitative and quantitative analysis of amino acids, protein, and carbohydrates.
In vitro studies- Good lab practices in BSL-II, maintenance of cell lines, sub-culturing, preservation, MTT assay, microscopy, lyophilization, and SEM.
Tumor studies in vivo- Induction of skin tumor, protectant administration, isolation of brain and liver, homogenate preparation, histology, biochemical estimations of ROS, LPO, GST, GPx, and LDH.
Plant tissue culture techniques- Micro-propagation and organogenesis.
