RESEARCH EXPERIENCE

Completed Masters Dissertation research under guidance of Prof. B.D Lakchaura, Prof. G.B. Pant University, Pant Nagar, India. My work was on Salmonella, pathogenic bacteria and involved isolation of this bacterium from the water samples collected from the Naini Lake of Nainital. This lake is a major water source for both humans and animals for the city itself and other suburban areas. The objective of the study was to assess the current status of Salmonella contamination in the Naini Lake and standardization of AP-PCR (Arbitrarily primed polymerase chain reaction) for molecular typing of isolates obtained from the water samples. The isolation technique involves both culture and DNA based detection techniques. The water sample was filtered through 0.4-micron filter paper, and then enriched in tetrathionate broth. The samples were screened for the presence of Salmonella by hisA gene targeted PCR. The samples showed positive result in hisA gene PCR and were streaked in Brilliant Green Agar. AP-PCR was used for molecular typing of the isolates obtained from the water samples. AP-PCR is also referred to as the randomly amplified polymorphic DNA assay employing a single short (typically 10 base pairs) primer that is not targeted to amplify any specific bacterial DNA sequence. The research work is examined by Doctors and professors from AIIMS, PGI in the final year viva.
