10. About Ph.D., thesis work

The Doctoral Thesis work deals with detection of emerging beta lactamase mediated

antibiotic resistance mechanisms in the clinical isolates of Acinetobacter spp. including

Extended spectrum beta lactamases (ESBLs), AmpC, Metallo beta lactamases (MBL), and

also the newer beta lactamases such as New Delhi metallo beta lactamase (NDM).

The current CLSI (Clinical and Laboratory Standards Institute) do not recommended

any phenotypic methods to detect ESBL mediated resistance mechanisms in Acinetobacter

spp. However, this method only recommended for E.coli and Klebsiella spp. In our

objectives, the CLSI recommended method was modified and applied for the detection of

ESBL detection in Acinetobacter spp.

Development of Multiplex PCR for the detection of ESBL mediated resistance genes

such as CTX-M, SHV, TEM and detection of genes which are responsible for the AmpC,

MBL, and NDM mediated antibiotic resistance mechanisms. Characterization of beta

lactamases antibiotic resistance genes. Finally correlate the phenotypic and genotypic

detection methods.

The clinical features were obtained from patients those infected with Acinetobacter

spp. and were significance was verified between beta lactamase producers and non-

producers.

Additionally, the biofilm formation and plasmid profiling and curing were performed

to see the significance with beta lactamase medicated resistance mechanisms
