TECHNICAL EXPERIENCES

Molecular biology techniques: Multiplex ligation-dependent probe amplification (MLPA) and quantitative PCR (qPCR) techniques to check frequency of homozygous and heterozygous CFHR1 and CFHR3 gene deletion in Hemolytic Uremic Syndrome (HUS) patients, their family members and healthy volunteers in Indian cohort. Preparation and use of Gold nanoparticles (AuNPs) in qPCR technique to increase their sensitivity. Western blotting to check complement factor H protein level. Total RNA isolation from eukaryotic cells and organs, cDNA synthesis and qPCR to studying kinetics of expression of various genes. Genomic and plasmid DNA isolation. Gene manipulating techniques like CFHR1 gene shDNA cloning in to pLKO.1-TCR cloning vector and transfection in to Hep3B hepatic cell line to prepare knockdown of CFHR1 gene and recombinant DNA techniques using overlap PCR for SP_0667 disrupted gene construction in S. pneumoniae strains using competence stimulating peptides (CSPs) for transformation.

Immunological techniques: Large Indian series of pediatric aHUS patients were analyzed for the presence of serum anti-CFH autoantibodies as well as CFH-anti-CFH immune complexes using ELISAs. Modified ELISA in presence of recombinant fragments of CFH and chemical (GuHCl, DEA and UREA) inhibitors determine the epitope specificity of anti-CFH autoantibodies, as well as of their relative affinity/avidity.

Animal handling: Experienced in handling of variety of mouse strains. Spleen, liver and brain isolation, Mice screening, retro-orbital blood collection and intra-peritoneal injection of NP40-Ficoll ((4-hydroxy-3- nitrophenyl) acetyl-Ficoll) for T-Independent response and NP11-ovalbumin for T dependent response. Determination of virulence of NCTC 7466 (D39) and ATCC BAA-334 (TIGR4) S. pneumoniae strains in BALB/c mice.

Cell culture: Culture of Hep3B hepatic cell line to prepare knockdown of CFHR1 gene. Human MCF-7 and HEK 293 cell lines cultured and expression of the transcription factor p53 and p21 gene were analyzed with the help of qPCR after treated with Paclitaxel (Taxol) drug. Handled variety of other cell lines also.

Bacterial culture: M.Sc. Dissertation gave me hands-on experience in bacterial culture, overlap PCR for disrupted gene construction, bacterial transformation & knockout mutant construction.

Flow cytometry Analysis: Performed FACS analysis to elucidate the role of Cisplatin and Vitamin C on apoptosis of SiHa cells. Also done FACS studies on JURKAT E6.1 cell line to evaluates the synergistic anticancer efficacy of bioactive Hydroxyapatite (HA) nanopar-ticles (HA NPs) loaded with Bendamustine HCl.

Molecular Diagnosis/ Diagnostic Cytogenetics: Gained Experience in Microarray based Diagnostic Cytogenetics and bioinformatics data analysis software (i.e. ChAS). Analyze and interpret data from microarray and sanger sequencing experiments which is used to detect the numerical and/or structural chromosome abnormalities. Associated congenital conditions, such as Down syndrome, Patau Syndrome, Edwards etc. Also employed in numerous other clinical situations, such as infertility, uniparental disomy (UPD), Parental (carrier) testing, maternal health etc. I have also done Cell free DNA isolation from blood and HRM-PCR.
