HANDS ON EXPERIENCE
1) "Dissertation Description "
Objectives:
. Collection and identification of AM fungi associated with medicinal
plants from Indian Thar Desert.
. To culture the AM isolates on some suitable hosts like cenchrus
ciliaris for mass multiplication and inoculums production
. To select the efficient AM strain for medicinal plants for its biomass
production and productivity.
. To evaluate influence of AM endophytes and various plants growth
promoting microorganism on various physiological and biochemical
changes in medicinal plants.
. To apply selected efficient AM strain and plant growth promoting
microorganisms for increasing biomass production of the medicinal
plants.
Research Methodology:
. Intensive periodical survey of the various regions of arid and semi
arid areas of Indian Thar Desert will be undertaken to collect and
describe AM endophytes associated with medicinal plants.
AM Endophytes:
. Root and soil samples was collected to recover AM fungal spores using
wet sieving and decanting technique of Gerdmann and Nicolson (1963)
. Anatomy of infected roots was described after suitable staining
(Phillips and Hayman).
. The percentage of root colonization calculated by grid line intersect
method (Giovannetti and Mosse, 1980), followed to isolate the local AM
fungal associates and establish them in pot culture.
Biochemical Analysis
. Peroxidase activity in the plant material was measured by
incubating the enzyme with guaiacol & hydrogen peroxide (Racusen &
Foote, 1965).
. Polyphenol oxidase activity will be measured at 420 nm, using the
method of mahadevan (1975).
Physiological Studies
. Soluble proteins in plant material determine by the comassive blue
method (Bradford, 1976).
. Total phenols in the plant material determine by using folin
ciocalteu's reagent (Bray and Thrope, 1954).
. Chlorophyll estimating of the plant material done by the method of
Lichenthaler & Wellburn (1983).
. Various nutrient viz phosphorus, nitrogen, etc estimate in the
plant material by the method of Jackson (1973).
. Total sugar and starch in the plant materials estimated by the
method of Dubius et al (1965).
2) "Microbiological Analysis of Different Pharmaceutical Samples"
