Project title :-“Isolation and characterization of potent oil degrading microbes

from oil contaminated sources and cloning and expression of oil degrading

genes in E.coli”.

Abstract :- The present study was carried out by production and characterization

biosurfactant producing microbes isolated from oil contaminated soils and

expression of biosurfactant producing genes in E.coli. soilsamples were collected

from different region of Lucknow and Kanpur which were then serilly diluted and

then streaked. Isolated cultures of oil degrading bacteria were characterized

according to Bergey’smannual. Small scale and large scale methods were used to

check the biosurfactant activity by using 2T oil and vegetable oil as substrate. The

best result were shown in 2T oil. Production of bacteria was done in its production

media to study the growth kinetics. Optimization of all the isolated bacteria was

done using suitable carbon sources, nitrogen sources, metal ions, and PH.

Biosurfactant activity of suitable bacteria was checked under optimized condition of

bateria. Plasmid isolation was done by alkaline denaturation methods and

transformation was performed by heat shock treatment in which genes are

expresssed from oil degrading microbes to non oil degrading microbes. Three

different cultures were isolated from different soil samples i.e, P.areuginosa,

B.cereus and Mycobacterium.
