PhD Research experience

> Underwent PhD Research at Department of Biochemistry, The Maharaja

Sayajirao University of Baroda, Vadodara, India

> PhD Research on "Physiological and Biochemical approach to improve

flowering and oil yield by phytohormone from Jatropha curcas" under the

guidance of Dr. Pushpa Robin

> The work mainly concerned with Jatropha curcas field study, Phytohormones

analysis, Enzymatic assay, DNA fragmentation study, Microscopy, Oil and

fatty acid analysis. Main theme of work is to study the role of

phytohormones, which increases flowering and fruit yield in Jatropha

curcas.

> Jatropha curcas flower and fruit development studied at field level.

Phytohormones such as Gibberellic acid, Auxin, Ethylene and ACC

(Aminocyclocarboxylate) were analyzed during inflorescences development

by GC-MS.

> Jatropha curcas seeds were analyzed for Oil extraction and fatty acid

analysis by Gas Chromatography.

> Enzymatic assay of Catalase, Superoxide dismutase and Peroxidase from

Abscission zone region during abscission/senescence of flowers and fruits

in Jatropha curcas, Hydrogen peroxide level was also study from

Abscission zone.

> DNA fragmentation study from abscission zone by DAPI dye and Gel

electrophoresis

> Cell death analysis by Light microscope and Transmission Electron

Microscope (TEM)

> In TEM, sample were prepared while exposed to osmium tetroxide and

glutarldehyde, a very thin sections were taken with the help of

Ultramicrotome and make grid by stained with silver staining. Sample was

prepared from fine region of Absicssion zone during the process of

senescence.
