Research Experience

2007 – 2012 As a Ph.D student

Project I: -AR-induced IL-6 via cAMP/Epac/PKC /P38 pathway

Our lab has done a lot of work on linking the sympathetic activation to cardiac inflammation during chronic heart failure

and has previously reported that β-adrenergic receptor (β-AR) induced IL-6 production in neonatal mouse cardiac

fibroblasts through Gs/cAMP/p38 pathway but independent of protein kinase A (PKA). My project focus on finding the

missing link between cAMP and p38 signaling pathways in cardiac fibroblasts. By using various protein kinase

inhibitors and adenovirus-mediated shRNAs to interfere with key signaling molecules, I reported β-AR activated

cAMP/Epac/PKC /p38 pathway to produce IL-6, and firstly demonstrated PKC is a novel downstream effector of

β-AR/cAMP/Epac pathway. (The paper has been accepted to British Journal of Pharmacology)

Project II: Gab1 (Grb-2-associated binder 1) protects the mouse heart against ischemia/reperfusion (I/R) injury

The project collaborates with Prof. Jincai Luo (Laboratory of Vascular Biology, Institute of Molecular Medicine, Peking

University, China), who generated the cardiac-specific Gab1 knockout mice. I independently do all the experiments to

monitor the progression of ischemia reperfusion injury of wild-type and Gab1 KO mice by echocardiography (Vevo 770TM),

hemodynamic measurements (via carotid using Millar catheter), 2,3,5-triphenyltetrazolium chloride staining after 30 min of

ischemia, 2 h or 24 h reperfusion. And I also involved in the isolation and culture of adult mouse cardiomyocytes. My

results show Gab1 deficiency dramatically increases cardiac I/R injury, which firstly present the critical role of Gab1 in I/R

injury. (All the data are summarized and submitted to Basic Research in Cardiology).
