Description:

POSITION SUMMARY:

The candidate will contribute to the evaluation and development of scalable AAV manufacturing technologies in support of efforts to develop human therapies. She/he will work to engineer a mammalian expression host as well as to help evaluate alternative expression systems such as baculovirus. The scientist will work to develop cell lines that be used in GMP manufacturing of AAV-based therapies.

ESSENTIAL FUNCTIONS:

The candidate will work to engineer a mammalian expression host for large-scale AAV production. This entails development of stable mammalian cell lines including transfection, selection, clone screening and cell banking. Screening will include assessment of transgene stability and AAV production upon scale-up and induction.

Strong molecular biology and cell culture experience is essential. Small scale AAV production and characterization in mammalian cell culture or Baculovirus systems for the assessment of manufacturing systems and/ or to support target validation activities is highly desirable.

Experience developing and characterizing stable mammalian cell lines is required. The candidate should have experience performing routine quantitative assays including qPCR, RT-PCR, Western Blot, ELISA and others.

The ability to generate and maintain experimental data in an electronic laboratory notebook in a highly organized manner is essential.
