Summary

Scientist with extensive expertise in analytical and physical chemistry, instrumentation, physics, polymer science, biochemistry, chromatography, mass spectrometry, drug discovery and screening including determination of protein-ligand binding characteristics (HPLC, LC-MS, other detection systems, e.g. RI, UV, MALS, ELSD, Viscometer). Significant experience in characterization and purification of small molecules and macromolecules (including proteins and peptides), their interaction, phase transactions, association and complex formation. Experience with measuring size of microparticles and their size distribution.

Managing experience as a group leader.

Objective: Position in Analytical Development

Experience:

Merck & Co., Cambridge, MA; Senior Principal Scientist 11/2009-1/2011

Developed a novel method of "Enhanced Ligand Affinity Characterization by LC-MS" (ELAC) to support ALIS (Automatic Ligand Identification System) operations and protein production in Protein Science Group. ELAC enables the complete characterization of the protein-ligand binding, including proteins and ligands solubility, stability and activity, Kd determination, stoichiometry, site of the binding and its type (covalent, non-covalent, specific, non-specific).

Schering-Plough Res Inst, Cambridge;

Sr. Principal Scientist/Group Leader 2/2005-11/2009

Developed synthesis, transferred to ALIS and filed a patent for novel media for drug screening process (Media-27). Developed method for ligand libraries preparation for ALIS screening that improves significantly the ligand solubility in aqueous buffer and prevents precipitation-mediated ligand loss. Developed method for Kd determination for GPCR targets.

Proposed ALIS-based method for quantitative bioanalytical analysis (including metabolic stability) and pharmacokinetical studies.

Developed, used and transferred to ALIS a specific system for high quality column packing (all SEC columns used by ALIS were packed using this system).

Neogenesis Pharmaceutical, Cambridge;

Principal Scientist/Group Leader 2/1998-2/2005

Was part of a team that developed a foundation for a model of the advanced drug screening process, ALIS.

Designed a special media for this process (Media 1 & Media 2).

Participated in the development of advanced LC-MS method for preparative ligands isolation.

Developed a novel three-column method for determination of equilibrium and kinetic binding characteristics, (Kd, Kon & Koff) for any target-ligand complexes.

Altus Biologics, Cambridge, MA; Senior Consultant 12/1996-2/1998

Developed a novel media based on cross-linked protein crystals and elaborated chiral

chromatography using the media.

Massachusetts Institute of Technology, Cambridge, MA;

Visiting Scientist 9/1996-12/1996

While working in the Department of Chemistry, developed a general thermodynamic model for protein solubility in organic solvents.

PerSeptive Biosystems, Framingham, MA; Senior Consultant 12/1995-8/1996

Developed methods for purification and analysis of different biological substances. Characterized porous media for perfusion chromatography using Malvern devices (for measuring particle size) and pore size distribution in the particles with using mercury porosimetry and a specific method of "Chromatographic Porosimetry".

Focal Interventional Therapeutics, Lexington, MA; Senior Scientist 9/1993-12/1995

Polymer, gel and drug characterization by a variety of analytical methods e.g., chromatography, light scattering, mass spectrometry, infrared spectroscopy, viscometry. Studied micelle formation from the "focal gel"; determined micelle size using multi-angle Light Scattering (MALS).

Wyatt Technology Corporation, Santa Barbara, CA; Senior Consultant 6/1993-9/1993

Application of multi-angle light scattering (MALS) for macromolecular characterization

Monsanto Chemical Co., Indian Orchard, MA;

Sr. Scientist/Group Manager 1990-1993

Managed a group of 7 people in Physical and Analytical Science Center . Developed analytical methods for characterization of linear and branched macromolecules, oligomers, block and statistical copolymers. Established a correlation between product properties on the molecular level and final product performance. Developed algorithm for production of SEC media with linear calibration according to molecular mass.

Purdue University, Indianapolis, IN; Visiting Professor 1989 -1990

Taught courses for graduate students (Department of Chemistry: Courses): Physical Chemistry, Properties of Macromolecules in Solutions; High Performance Liquid Chromatography (HPLC), and Instrumental Analysis.

Institute of Macromolecular Compounds, Leningrad, USSR; Scientist, Senior Scientist,

then Principal Scientist 1974-1988

Developed a theory and methods for study of macromolecules in solution, their phase

transactions, interaction, association and complex formation. Developed a theory of

macromolecular chromatography and theory for chromatographic process optimization (see "Modern Liquid Chromatography of Macromolecules", Elsevier, 1984). Developed a variety of analytical methods using SEC, RPC, NPC, IEC, chiral LC and light scattering for separation, purification and characterization of proteins, small molecules, oligomers and polymers.
