JOB PROFILE

Molecular Biology: Cloning and expression of heterologous proteins in Bacteria, Yeast and Mammalian expression systems. Standardization of PCR and RT-PCR, Assembly of genes using SOE PCR method, DNA sequencing, Genomic DNA isolation (From different sources), Total RNA isolation and Plasmid DNA isolation. Downstream process purification of recombinant proteins using chromatographic techniques.

HPV prophylactic vaccine:

• Cloning and expression of HPV L1 (16L1,18L1,31L1,45L1 and 52L1) capsid proteins in

Pichia pastoris. Assembled VLPs purified by affinity chromatography. Immuno efficacy trails with bi and pentavalent VLP formulation in mice.

• Tandem cloning of HPV16 L1 and L2 in pichia pastoris.

• Cloning and expression of HPV L1 (16L1 and 18L1) in Salmonella Typhi Ty21a.

Transnasal immunization followed by CMI response in mice was evaluated by cell based

assays.

• Cloning of HPV genes in mammalian expression systems.

• Periplasmic expression of HPV proteins in Bacteria.

• Preparation of HPV Pseudovirions for determining the sera titer in vaccine efficacy studies.

Coccidial Vaccine: Expression of Coccidial proteins (EtMIC-1 and EtMIC-2) in tobacco by

Agro infiltration.

Microbiolgy: Bacterial strains like DH5α, BL21 and Salmonella isolation propagation and maintenance, biochemical characterization and pure culture development.

cGMP Seed Banks: Preparation of Recombinant HPV salmonella 16L1 and 18L1 master and working banks and played a vital role in technology transfer to production department.

Virology : Isolation of viruses (Bovine herpesvirus -1) from clinical samples, adaptation of virus isolates to in vitro mammalian cell culture, propagation, plaque assay, purification, TCID50, filtration/concentration, characterization of viruses (BHV-1) for vaccine development, , virus bank preparation.

IBR Vaccine: Evaluation of inactivated oil and Al-gel BHV-1 vaccine’s against Bovine herpesvirus-1 (BHV-1) in cattle. Studies on Humoral and Cell mediated immune response in IBR vaccinated cattle by ELISA, MNT, FACS and ELISPOT.

Cell culture : Maintenance and characterization of mammalian cell lines (BHK-21, VERO, MDBK, MDCK, MRC5,293TT etc.,) Isolation and cryo-preservation of PBMC’s and Splenocytes from vaccinated and natural infected animal for the study of cell mediated immune response (Cytokine Profile) against different antigen stimulants.
