Analysis of Mammary Gland Transcriptome and Proteome for Identification of Probable Mammary

Biomarkers:

Mammary gland is made up of a branching network of ducts that end in alveoli. Terminally differentiated mammary

epithelial cells (MECs) constitute the innermost layer of aveoli. They are milk-secreting cuboidal cells that secrete

milk proteins during lactation. Little is known about the expression profile of proteins in the metabolically active

MECs during lactation or their functional role in the lactation process. In the present investigation, we have reported

the proteome map of MECs in lactating cows using 2DE MALDI-TOF/TOF MS and 1D-Gel-LC-MS/MS. MECs

were isolated from milk using immunomagnetic beads and confirmed by RT-PCR and Western blotting. The 1D-Gel-

LC-MS/MS and 2DE-MS/MS based approaches led to identification of 431 and 134 proteins, respectively, with a total

of 497 unique proteins. Proteins identified in this study were clustered into functional groups using bioinformatics

tools. Pathway analysis of the identified proteins revealed 28 pathways (p < 0.05) providing evidence for involvement

of various proteins in lactation function. This study further provides experimental evidence for the presence of many

proteins that have been predicted in annotated bovine genome. The data generated further provide a set of bovine

MEC-specific proteins that will help the researchers to understand the molecular events taking place during lactation .

Study of Antimycotic potential of Nitrogen and Sulpher containing Heterocycles against Aspergillus Species

with Proteomics Approach:

We developed a new and useful one-pot synthetic protocol for diverse 2-thioxo-4(3H) -quinazolinones (through

benzotriazole methodology) by DBU catalyzed addition of anthranilic acid/ester and amines to bis-benzotriazolyl)-

methanethione. The method is convenient, simple in handling, and good to high yielding. the result of anti-Aspergillus

evolution reveled that one of the 2-thioxo-2, 3-dihydroquinazolin-4(1H)-ones derivative i.e. 3-(4-Phenyl-thiazol-2-yl)-2-

thioxo-2,3-dihydro-1H-quinazolin-4-one (DDRVKT-4Q) and the compound DDRVKT-4Q exhibited appreciable activity

. The potency of compound was found concentration of 3.125 µg/disc of DDRVKT-4Q produced a diameter of zone of

inhibition equivalent to that produced by 1.25 µg/disc of amphotericin B. A concentration of compound up to 625 µg/ml

was found to be completely non-toxic to the cells. The fungicidal activity of the compound DDRVKT-4Q was caused

99.8% of loss of viability of spore after 8.5 h of incubation; however Amp B caused 99.8 % loss of viability of spore

after 4.5 h. The cytotoxicity of the compound DDRVKT-4Q was found to be non toxic at concentration 625 µg/ml. and

its lysed only 35.9% of human erythrocytes, at the highest dose tested, Amp. B was found to be lethal to all cells at 39.05

µg/ml. Attempts were made to identify the possible protein target of the compound in the pathogen. It was observed that

the treatment of pathogen with DDRVKT-4Q targeted the expression of four proteins having molecular weights 18 kDa

37 KDa and 43 KDa proteins was completely inhibited or down regulated by the compound the extra cellular. The novel

compound 3-(4-Phenyl-thiazol-2-yl)-2-thioxo-2, 3-dihydro-1H-quinazolin-4-one, having antifungal activity can be

exploited further to develop new ideal antimycotic drugs.
