proctoring exams, meeting with students after hours to review material,

Cystic Fibrosis Research in the Department of Physiology

. Participated in the design and development of a genetic screening assay

utilizing multiplex PCR for the diagnosis of 18 SNP mutations in the CFTR

gene that are prominent within the Mexican population.

. In an effort to maximize stringency and eliminate false-positives, we

introduced a novel Pyrimidine mismatch three nucleotides in from the 3'

end of the oligonucleotide primers and also customized the 10X PCR

reaction buffers for specific sets of primers.

. Site-directed mutagenesis was utilized to provide mutant DNA for testing

purposes.

. In a parallel project to increase the processivity of the thermostable

polymerase used in the multiplex, we constructed a novel method to

chelate divalent cations to one half of the polymerase active sites. This

reduced competition for these ions by other components in the reaction

cocktail and increased the concentration of DNA replicated from PCR.
