Summary of Qualifications

Research Scientist with several years of research experience in cell biology and immunology with a specialization in development of cellular products for immunotherapy in a clinical research setting. Demonstrated expertise and technical skills to plan and execute experiments to identify and generate suitable cellular products for immunotherapy. Well experienced in the process to generate and characterize virus specific and leukemia specific cytotoxic lymphocytes (CTLs), magnetic beads and antibody based enrichment, isolation using FACS sorter, multi-color FACS to characterize, large-scale expansion of cellular products in GMP environment, cryopreservation, development and optimization of FACS panels to cell sub-set identification, CTL assay and ELISPOT assays to characterize and check the cytotoxicity, multiple cytokines assay using bioplex and gene expression analysis using qPCR. Performed immunohistochemistry, radio labeling of peptides and purification, radio immuno assay (RIA), and ELISA extensively for my research as well as for a major clinical research. Hands on experience in mammalian primary cell culture, EBV-LCL transformation, cell proliferation assay, immune tolerance assay, characterization of large molecules (Proteins) by gel electrophoresis and western blot analysis, cell-based assays, and biochemical assays. Prepared and implemented SOPs and protocols. Experienced in good time, project and lab management skills. Excellent written and oral communications, troubleshooting, and collaborating with cross-functional teams. Strong knowledge in current Good Manufacturing Practice (cGMP) and Good Laboratory Practice (GLP) and Good documentation practice.

Five years of clinical research experience gained from two multicenter clinical trials, which investigated the immune reconstitution after cord blood transplantation; I was the laboratory contact for the coordinators and responsible to receive samples, prepare samples for multiple experiments in aseptic condition which includes, FACS staining, acquisition & analysis of data, set-up cell culture experiments to study the antigen specific (virus) T cell proliferation, immune tolerance and quantification of multiple cytokines using bioplex, magnetic beads based separation analysis of data and report to PI, cryopreservation of cellular subsets and shipment to other centers for further analysis.

Managerial Skills

* Demonstrated independent thought/creativity in science and scientific techniques.

* Excellent communication (oral and written) and multitasking skills

* Worked well independently and in a multi-disciplinary team environment.

* Maintained appropriate notebook records of all studies (Good documentation practice), and satisfied all SOP lab requirements.

* Maintained laboratory in compliance with the Bio/chemical/radioactive lab safety guidelines.

* Computer Skills: MS-Word, Excel, Power Point and Bioinformatic tools for analysis.

Professional Experience

2011- present

Division of Blood and Marrow Transplantation and Cellular Therapies, Children’s Hospital of Pittsburgh

Pittsburgh, PA

Research Associate Sr.

Designed and developed methods to generate and characterize effective leukemia and virus specific cytotoxic T lympocytes from expanded cord blood T cells using IL15.

Demonstrated that the cytotoxicity enriched among CD137 positive subset and thus the CD137 expression can be used as a marker to isolate and expand antigen specific CTLs.

Laboratory contact and responsible for two clinical trials, which investigated the immune reconstitution after cord blood transplantation (experiment setup, data acquisition maintenance and analysis).

2008-2011

Pediatric Blood and Marrow Transplant Program, Duke University Medical Center

Durham, NC

Research Scientist Sr.

Expanded in large scale and characterized the cord blood T cells in GMP environment for a planned clinical trial to adoptively infuse the expanded CBT cells to patients after cord blood transplantation (submited to FDA as investigational new drug application).

Set-up experiments and analyzed the data to study the immune re-constitution and immune-tolerance after cord blood transplantation for two clinical trials.

2005-2008

Department of Cell and Developmental Biology, University of North Carolina

Chapel Hill, NC

Research Assistant Professor

Designed and executed experiments to study the role of androgen binding protein (ABP) on intra-testicular testosterone level and gene expression using Affymetrix mouse genome array using RNA from the testis of a transgenic mice which has over-expressed ABP.

Provided technical expertise to the ImmunoAnalysis Core Laboratory at the Lineberger Comprehensive Cancer Center to quantify the proliferative and apoptotic index from prostate biopsies for a major clinical research project, which evaluated the racial difference in prostate cancer between African American and Caucasian American men.

2004-2005

Department of Cell and Developmental Biology, University of North Carolina

Chapel Hill, NC

Postdoctoral Research Associate

Explored the role of androgen binding protein on intra-testicular testosterone levels using ABP-transgenic mice

1999-2004

Department of Cell and Developmental Biology, University of North Carolina

Chapel Hill, NC

Postdoctoral Fellow

Demonstrated the role of androgen binding protein on testicular germ cell proliferation and apoptosis using ABP-TG mice as a model.

Created a reference database describing the proliferative and apoptotic index for the germ

cell development using flowcytometry as a tool.

1997-1999

Department of Reproductive Biology, All India Institute of Medical Sciences

New Delhi, India

Research Associate

Experimentally induced BPH in rhesus monkeys using steroids to develop an in vivo model.

Developed an in vitro primary culture system for prostate epithelial cells.

1995

Hi-tech Diagnostic Center

Chennai, India

Part-time Technologist

Managed the RIA laboratory - assay different hormones, analyze and report to the Director.

Education:

PhD, Endocrinology, University of Madras, India

Awards & Honors:

* Won the third prize for the best research article published in Reproductive Biology & Endocrinology Journal during the year 2005.

* John E. Fogarty Fellowship (1999-2004) from the National Institutes of Health (NIH), USA.

* Senior Research Fellowship (1992-1994) from the Council for Scientific and Industrial Research (CSIR), Government of India.

* University Research Fellowship (1990-1991) from the University of Madras, India.
